Damnoen Saduak runs daily, but boats and vendors thin out well before lunch. Amphawa is a Friday–Sunday afternoon-and-evening market. Taling Chan and Khlong Lat Mayom run on weekends (and some public holidays) only.

What to expect on the day
Pickup is around 07:00 because the schedule only works in one direction: Maeklong Railway Market first or Damnoen first, timed to catch a train pass — the whole market folds its umbrellas back for a few slow seconds of passing locomotive (the published timetable has trains arriving roughly 08:30, 11:10, 14:30 and 17:40 — the railway does change it, so confirm on the day). It's genuinely surreal and costs nothing beyond the tour.
At Damnoen Saduak you'll be offered the paddle-boat ride — 2026 reports put a shared boat at roughly ฿150–300 a head, there's no official rate card, and many tours leave it to you, so check yours. Take it once; walking the banks shows you only backs of heads. Boat vendors sell boat noodles, mango sticky rice and coconut ice cream at visitor prices. Haggle with a smile; opening prices assume you won't.
The two honest caveats: you'll spend roughly 3 of the 7.5 hours on a bus, and from about 09:30 the main canal is a floating traffic jam of tour groups photographing each other. On the published timetable, trains arrive at Maeklong roughly at 08:30, 11:10, 14:30 and 17:40, and the market folds itself away and rebuilds in about a minute each time. The State Railway does change the timetable, so treat those as a guide and confirm on the day. Morning combo tours are built around the 08:30 or 11:10 pass — check which one your tour catches before booking.
